Over the entrance to Mrs. Who's (Mindy Kaling's) house is a street-number sign with the eight hanging lopsided, forming an infinity symbol.

In the book, Meg and her family are Caucasian, and in the movie, the Murry family is multiracial. This became controversial among fans of the book and its sequels, but filmmakers believed a multiracial family would be more relevant and relatable for contemporary audiences. Chris Pine has said that the multiracial family dynamic was actually one of the aspects in the script which attracted him to the project.

Following mixed-to-negative critical reactions and a disappointing box office in the U.S., the theatrical release of the film was cancelled in several overseas countries such as Belgium and Netherlands, going straight to Video-on-Demand instead.

In the book, the Murry family includes identical twin brothers Sandy and Dennys, who do not appear in the movie.

Oprah Winfrey got the role on the spot without auditioning. She was texting director Ava DuVernay one day and DuVernay mentioned that she was developing a new "A Wrinkle in Time adaption" and asked if she could work with her, and Oprah said "no problem".
