This film repeatedly makes fun of Hallmark movies. Luke Macfarlane (Aaron) has starred in 14 Hallmark movies.

Shortly after its release, a theater chain told Universal Studios that it wanted to pull the trailer because of the gay content; Universal convinced it not to.

Luke Macfarlane revealed he had no inhibitions during his raunchy nude sex scenes with Billy Eichner. "Specifically we had an LGBT intimacy coordinator, so they were familiar with language that might be a little bit different. Billy and I felt really comfortable. We gave each other permission to go for it." He said there was only one thing he said no to: "There was a moment when Billy was like should we spit on each other? And I was like, 'Nope! Nope!' So that was something that I decided not to do."

After the film's underperformance at the box office (it cost around $22 million to make but grossed a mere $4.8 million over its opening weekend, less than half of Universal's projected $10 million), writer/lead Billy Eichner tweeted his disappointment on Oct. 8, 2022, saying, "That's just the world we live in, unfortunately. Even with glowing reviews, great Rotten Tomatoes scores, an A CinemaScore etc, straight people, especially in certain parts of the country, just didn't show up for Bros. And that's disappointing but it is what it is." According to director Nicholas Stoller, exit data for the film found its audience to be overwhelmingly gay and bisexual men.

Queer Eye (2018) is one of the pieces of pop culture that gets lampooned in this movie. Jai Rodriguez, who plays Jason (Bobby's brother), was an original cast member of the first, early-2000s incarnation of that reality show, Queer Eye (2003) (original title: "Queer Eye for the Straight Guy".
